چکیده
The polyamine spermidine (NH,(CH&NH(CH&NH,) is widespread in nature. It is present in mammalian tissues (1, 2), plants (3), bacteria (47), ribosomes (6, 8), and bacteriophages (9-11). There are various methods for its identification, including steam distillation (12)) paper chromatography (13-18)) paper electrophoresis (14, 16, 17, 19), ion exchange chromatography (2, 5), gas chromatography (31), and bioassay (20). Paper chromatography, electrophoresis, and steam distillation are not precise methods. Ion exchange chromatography, on the other hand, is accurate but too elaborate for routine use. This paper deals with a specific and sensitive enzymatic method for the determination of spermidine. The method is based on the formation of A’-pyrroline from spermidine, by freeze-dried spermidine-adapted Xerratia marcescens cells. The enzymatic oxidation of spermidine proceeds according to Reaction 1 (2123).

متن کاملA new simple enzymatic assay method for urinary polyamines in humans.
We developed a new simple enzymatic assay method for measuring urinary polyamines (total amount of putrescine, spermidine, and cadaverine), using an acylpolyamine amidohydrolase and a putrescine oxidase. First conjugated polyamines (putrescine, spermidine, and cadaverine) in urine were hydrolyzed by incubation with an acylpolyamine amidohydrolase at 30 degrees for 1 hr. then, free polyamines we...

متن کاملThe non-enzymatic hydrolysis of oligoribonucleotides VI. The role of biogenic polyamines.
Single-stranded oligoribonucleotides containing UA and CA phosphodiester bonds can be hydrolyzed specifically under non-enzymatic conditions in the presence of spermidine, a biogenic amine found in a wide variety of organisms. In the present study, the rate of oligonucleotide and tRNA(i)(Met)hydrolysis was measured in the presence of spermidine and other biogenic amines. It was found that sperm...
متن کاملSpermidine but not spermine is essential for hypusine biosynthesis and growth in Saccharomyces cerevisiae: spermine is converted to spermidine in vivo by the FMS1-amine oxidase.
In our earlier work we showed that either spermidine or spermine could support the growth of spe2Delta or spe3Delta polyamine-requiring mutants, but it was unclear whether the cells had a specific requirement for either of these amines. In the current work, we demonstrate that spermidine is specifically required for the growth of Saccharomyces cerevisiae. We were able to show this specificity b...
